ABSTRACT
The data emerging from some of the cancer registries in sub-Saharan Africa indicate that the burden of cancer is increasing rapidly. Life style, environmental, dietary and chemical factors have all been suspected to play significant roles in the occurrence of malignancies. Rifampicin which is a first line antituberculous agent has been identified as a class three mutagen by the International Agency for Research on Cancer (IARC). Genotoxic investigation of both environmental pollutants and therapeutic agents were carried out using both modified Ames assay and Allium cepa (Linn) model. Instead of the Salmonella typhimurium TA 100 or TA 98 used by Bruce Ames, E.coli 0157: H7 was used in the modified Ames assay. Alteration of at least three biochemical characteristics of the normal biochemical characteristics of this organism (lactose and fructose fermenting, indole positive, urease negative, citrate negative, motile and gas production) was taken as bench mark for mutagenicity in this study.
DNA extraction and digestion with restriction enzymes were done using the standard method. Allium cepa assay was done to determine the mitotic index, root growth and chromosomal aberration using the standard method. Modulatory effect of antioxidants on rifampicin toxicity was done with rats using sub-chronic toxicity method and the modulatory roles of antioxidants on rifampicin mutagenicity was done using micronucleus assay.
